Accurate Drug Reposition through Non-Tissue Specific Core Signatures from Cancer Transcriptomes

Description

We used TCGA cancer transcriptomes to generate non-tissue specific core signatures (CSs) of human coding gene mutaitons. Then we used the CSs to scan Connectivity Map (CMap) drug profiles to generate d2gNES and d2gCSI values. A positive d2gNES indicates a drug treatment profile is with the same expression direction of a gene mutation, and a negative d2gNES indicates the opposite direction to a gene mutation. The d2gCSI ranges from 0 to 1, indicating the specificity of a drug targeting a gene.
