<dc:title>Codes for Higher actin contractility and cargo-loaded clathrin pits create midplane-peaked apico-basal tension profiles in HeLa cells </dc:title>
    <dc:publisher>Mendeley Data</dc:publisher>
<dc:description>Folder “actin_myosin_or_ezrin_analysis” contains the codes for the analysis of the ezrin-actin or myosin-actin colocalization at the cortex of the cell and the code to collect the data from the .mat files obtained from each cell analysis.

Folder “flipTR_lifetime_analysis” contains the code for the flipTR lifetime analysis at the periphery region of the cell (“boundarycellforTanmoy.m”), and for PMS (“lifetime_analysis_pms.m”), from which the lifetime value was collected at the membrane, and then analysed. We also have the code “Tanmoy_flipprofile_collate.m” to collate the flipperTR profile data from each profile in the main code. Also, we have code “section_visualing.m” to visualise the surface of the membrane overall along the height of the cell, to understand if there is a prevailing tension or order gradient along the cells’ or PMS height.

Folder "for_plotting" contains a program that visualises data by dividing the dataset into equal segments and calculating the median and error for the plot, creating a profile of scatter data from multiple cells.

Folder “OT_image_analysis” contains two folders, the first “OT_image_analysis”, used for the force measurements by image analysis, and the other is “Stage_Run_Labview_code”, used to move the stage at a particular velocity of 0.5 μm/sec away from the trapped bead for tether pulling experiments. The “OT_image_analysis” module contains the required functions and code to determine the centroid of the bead, which is optically trapped for force measurements. The main code here is “trap_all_files.m” used to find the centroid of the bead by fitting the periphery to a circle.

Folder “transferrin_clathrin_analysis” has a code “tf_clathrin_cortex_wc_analysis.m” to analyse transferrin and clathrin colocalization at the cortex per image stack. After generating it, the data was collected by using the code “data_collect_all.m” for further analysis.
