Pacemaker neurons exert control over neuronal circuit function by their intrinsic ability to generate rhythmic bursts of action potential. Recent work has identified rhythmic gut contractions in human, mice and hydra to be dependent on both neurons and the resident microbiota. However, little is known about the evolutionary origin of these neurons and their interaction with microbes. In this study, we identified and functionally characterized prototypical ANO/SCN/TRPM ion channel expressing pacemaker cells in the basal metazoan Hydra by using a combination of single-cell transcriptomics, immunochemistry, and functional experiments. Unexpectedly, these prototypical pacemaker neurons express a rich set of immune-related genes mediating their interaction with the microbial environment. Functional experiments validated a model of the evolutionary emergence of pacemaker cells as neurons using components of innate immunity to interact with the microbial environment and ion channels to generate rhythmic contractions. Time series single cell RNA sequencing analysis of smooth muscle cell (SMC) to mesenchymal cell transition process in TGFβR2iSMC-Apoe-/- and Apoe-/- mice. Characterization of TGFβR2iSMC-Apoe-/- mice after 1 month of high cholesterol high fat by histology, imaging mass cytometry (IMC), histoCAT, CyTOF, and immunocytochemistry. Aortic smooth muscle cells from TGFβR2iSMC-Apoe-/- express stem cell markers (CD105, CD73, CD90, Sca-1, CD44), mesenchymal cell markers (Osteopontin, Aggrecan, Adiponectin). To show clonal origin of smooth muscle cell-derived mesenchymal cells in TGFβR2iSMC-Apoe mice-/-, we replaced mTmG reporter strain TGFβR2iSMC-Apoe mice generating Myh11CreERT2;Tgfbr2f/f;Apoe-/-;Confettif/f and Myh11CreERT2;Apoe-/-:Confettif/f (control) lines.

The objective of this study was the evaluation of the outcome of a special interdisciplinary therapy program depending on the beginning of the rehabilitation. The outcome of a specialized hand rehabilitation immediately after as well as short-term follow-up of 76 patients with injuries of the hand respectively wrist and CRPS was analysed with the use of self-assessed parameters and objective functional scores. The patients were divided into a group with an early (< 120 days after trauma) or late beginning of the rehabilitation. Better results for the early beginners were shown with statistical significance for the DASH–Score as well as ROM at follow-up. Especially patients with CRPS and finger injuries had a benefit. The study shows the importance of an early beginning especially with respect to function. There are differences in between different injuries or diseases with respect to these effects, especially patients with CRPS seem to profit.
