The data describe the preparation, optimization, characterization, and evaluation of nanostructured lipid carriers (NLCs) loaded with ondansetron hydrochloride (OSH), a water-soluble drug. A modified cold high-pressure homogenization method was used, and the pH-dependent solubility of OSH was exploited to obtain NLCs with the enhanced entrapment efficiency (EE). The optimized formulation exhibited the sustained-release characteristic of the drug in vitro. After single subcutaneous injection in rats, the optimized OSH-loaded NLCs could prolong the drug release until 96 h with an extended mean resident time and enhanced systemic exposure compared to the OSH solution.
