Description

This retrospective case–control study analyzed 137 ALL patients and 350 ethnically-matched controls using six‑digit NGS typing of HLA‑A, ‑B, ‑C, ‑DRB1, ‑DQA1, ‑DQB1, and ‑DPB1 loci. NGS-based high-resolution typing identified novel, population-specific HLA associations with ALL in ethnic Kazakhs, with class II effects predominating and additional class I signals at HLA-B and HLA-C.

Steps to reproduce

HLA typing for both ALL cases and controls was performed at the same certified facility (Scientific and Production Center for Transfusiology; Astana, Kazakhstan), using the same workflow and setup. Cases and controls were genotyed using the MiSeq sequencing platform, the library preparation method, and the HLA Twin™ v4.9.0 bioinformatics pipeline. Peripheral blood samples were collected in EDTA-K₂ tubes, and genomic DNA was extracted by the automated BEX12 system (Inno-Train Diagnostics, Germany) using magnetic particle-based separation.
