Included are full length western blots that accompany the publication "Impaired autophagic-lysosomal fusion in Parkinson's patient midbrain neurons occurs through loss of ykt6 and is rescued by farnesyltransferase inhibition" in the Journal of Neuroscience 2023 PMID: 36788031 DOI: 10.1523/JNEUROSCI.0610-22.2023 ). Our imumunodetection system includes the use of two secondary antibodies (anti-mouse or anti-rabbit IgG) that are conjugated to two different fluorophores and are detected on either the 700nm channel (ch) or 800nm channel, simultaneously on the same membrane. Some membranes appear as high exposures (exp) or low exposures, which refers to differences in scanning intensity and contrast settings. Throughout the data set, all samples that were intended to be compared directly (treated vs untreated, mutant vs. healthy control vs. isogenic corrected) appear on the same blot and loaded as close as possible for accurate quantitative comparisons and to account for differences in the blot backgrounds. CBB refers to Coomassie Brilliant Blue staining. The CBB stained gel that appears under the western blot is the proteins remaining in the gel after the transfer protocol, and corresponds to that same blotted membrane.
