Dataset on cytotoxicity of activated and expanded natural killer cells against multiple myeloma side population cells: side population analysis

Description

Two different myeloma cell lines (L-363 and OPM-2) and three different myeloma samples at different disease evolution stages (smoldering myeloma, multiple myeloma and plasma cell leukemia) were analyzed searching for side population. Side population could constitute the stem cell compartment in multiple myeloma as this population exhibits drug-resistance. We decided to perform RNA-seq experiments with purified side population (SP) and non-side population (Non-SP) from the different samples to analyze if the SP could over-express genes involved in the stemness of this population compared to the non-SP. Also, we aimed to find over-expression of some potential markers of stemness that have been described for another diseases, whereas there is no evidence of specifics markers in multiple myeloma. Gene Ontology Analysis and Gene Set Analysis were performed comparing SP vs. non-SP. We did not find any difference in the known stem cell markers for other diseases. However, GSA allowed us to verify that SP cells expressed CD138 (corroborating what we found by flow cytometry). Gene Ontology analysis evidenced the over-expression of genes involved in the asymmetric division, characteristic of stem cells . For GSA, a false discovery rate (FDR) < 0.01 and 2-fold change were used to consider relevant differences. Enrichment analysis was performed based on Gene Ontology (GO) Consortium. GO terms with a fold change > 2 and p < 0.05 were considered functionally relevant.
