Along an 837 km transect in the eastern Qilian Mountains, eight typical sites dominated by S. chamaejasme were selected. Within each site, five healthy plants were randomly chosen . Soil samples designated as the 'root-surrounding soil' were collected from beneath each plant at a designated distance of 5 cm from the root surface. The five soil samples collected per site were then pooled in equal mass to form one composite soil sample. Following this method, three composite soil samples were collected per site. Samples were transported to the laboratory under low-temperature conditions. Upon arrival, impurities were removed, and the soil was sieved through a 2 mm mesh and homogenized. One portion was air-dried for physicochemical analysis, another portion was flash-frozen in liquid nitrogen and stored at -80 °C for DNA extraction, and the remainder was stored at 4 °C.
