In the first library, nuclei were not fixed and processed directly for subsequent reactions. In the second library, nuclei were fixed and then frozen in liquid nitrogen. In the third library, nuclei were permeabilized right after fixation and then frozen in liquid nitrogen. The three libraries were generated as pilot data with a small split-pool barcoding scale. The three libraries were sequenced at a low depth with the spike-in sequencing approach at Center for Genome Sciences & Systems Biology at Washington University in St. Louis.
