Proteomic dataset of cryopreserved sperm from Nelore bulls (Bos indicus)
Description
The objective of this study was to characterize the proteome of cryopreserved sperm from Nelore bulls (Bos indicus). Proteomic results were correlated with sperm kinematic subpopulations. The data is organized into individual files corresponding to each bull, containing protein identification and quantification data from thawed sperm samples.

Steps to reproduce
Semen samples were collected from bulls (n = 10), and aliquots containing 50 - 60 × 10⁶ sperm were processed for proteomics. Samples were washed with PBS (pH 7.2) to remove extender and after the last washing the supernatant was removed and added RIPA buffer supplemented with protease inhibitors to perform the protein extraction. Protein was submitted to SDS-PAGE following by in-gel digestion, and nLC ESI-MS/MS using an Orbitrap Fusion Lumos Tribid mass spectrometer. Proteins were identified by database searching and relatively quantified by spectral counting using the exponentially modified protein abundance index (emPAI). Proteomic data was analyzed using MetaboAnalyst 6.0 (https://www.metaboanalyst.ca/), and Spearman correlation was performed using R and RStudio.
