Drosophila embryos were collected and subjected to two-colour FISH, highlighting cytogenetic positions 37B and 89E. 3D stacks were collected from 3-5 different embryos. Optical sections were collected at 0.5 μm intervals along Z-axis using a Leica SP8-UV microscope or a Zeiss AXIO imager. Relative 3D distances between FISH signals were analyzed in approximately 80 to 120 nuclei per 3D stack using the Imaris software (Oxford Instruments).
