To investigate the response of rhizobacteria communities to soil suspension under different phosphate conditions, rhizosphere soil samples from Col-0 and fer-4 grown in HP-T-SS or LP-T-SS were collected for soil experiment 16S rRNA gene amplicon sequencing. Soil samples from HP-T-SS (without plant) were collected as control and considered as bulk soil (BS).

Steps to reproduce

Total genomic DNA was extracted using PowerSoil® DNA Isolation Kit (Mobio, America) following the manufacturer’s instructions. The bacterial 16 S rRNA gene V3-V4 region was amplified using universal primers 343 F: 5’-TACGGRAGGCAGCAG-3’ and 798 R: 5’-AGGGTATCTAATCCT-3’. After being purified with the AMPure XP beads (Agencourt, America) twice, equal amounts of purified amplicon were pooled for subsequent sequencing at Shanghai OE Biotech. Co., Ltd. (Shanghai, China) using the Illumina MiSeq platform. Raw sequencing data which was in FASTQ format was processed by removing, splicing, and denoising. All reads obtained from twenty samples were divided into OTUs.
