Rhinoceros Serum MicroRNAs

Description

MiRNA sequences and counts in serum of four species of rhinoceros: black (Diceros bicornis), greater one-horned (Rhinoceros unicornis), Sumatran (Dicerorhinus sumatrensis) and white (Ceratotherium simum). Serum samples from black and Sumatran rhinos were from individuals categorized as healthy, unhealthy (afflicted with disease other than iron overload), and afflicted with iron overload disorder. Serum samples from greater one-horned and white rhinos were from healthy individuals. RT-qPCR Raw Cq values for serum miRNAs are also included for additional serum samples collected from black and Sumatran rhinos. This excel file contains three tabs: sample info, sequencing data, and PCR data. A few of the samples overlap between the datasets - refer to assigned sample number and animal ID to identify overlaps. Version 2: PCR data was added to file Version 3: Sequencing files and PCR files were combined into one excel file for ease of use and additional sample demographic info was added. Version 4: Sample info was combined into one tab.

Steps to reproduce

Serum samples (n = 26; 1 -3 samples/individual) from individuals of four rhinoceros species maintained at North American institutions were selected for miRNA sequencing: black rhinos (Diceros bicornis; n = 5 individuals), greater one-horned rhinos (Rhinoceros unicornis; n = 4 individuals), Sumatran rhinos (Dicerorhinus sumatrensis; n = 4), and white rhinos (Ceratotherium simum; n = 4). Samples had been previously banked during veterinary procedures and were used for this study opportunistically. The samples were shipped to LCSciences (Houston, TX) for miRNA sequencing and bioinformatics analysis. Sequences will be submitted for inclusion in the miRBase database, at which point miRNA names may change. Specific miRNAs (n = 7) were selected for RT-PCR analysis of serum samples from black and Sumatran rhino samples (n =99) to evaluate the potential of 5 specific miRNAs (bta-miR-143_R-2; bta-miR-16b_R+1; mmu-miR-146a-5p_R+1; bta-let-7g_R+1; and bta-miR-30e-5p) as biomarkers for in Iron Overload Disorder (IOD) in these two rhino species. The other two miRNAs (bta-let-7c-5p and bta-miR-331-5p_R+1) included were used as housekeepers.
