In vitro biological effects of polysaccharide (funoran) from Gelidiopsis variabilis on fibroblasts, keratinocytes, macrophages, and colorectal adenocarcinoma cells
Description
Dataset 1: Cytotoxicity, metabolic activity, and SOD activity of Gelidiopsis variabilis (GV) polysaccharide fractions (Funoran) in human dermal fibroblasts (HDF) Description: This dataset comprises raw data from three independent experiments evaluating the effects of six Funoran fractions (GV-1A, GV-1B, GV-2A, GV-2B, GV-3A, and GV-3B) on primary human dermal fibroblasts (HDF). HDF: MTT (1.00–0.02 µg/µL, 7 dilutions, 24 h) and SOD (0.5 µg/µL, 24 h): normalized to NT, PC = mitomycin C, Dunnett's test. alamarBlue (0.5 µg/µL, 24–72 h): Šídák's test vs. NT. All data: mean ± SEM (n=4). SOD activity in U/mg protein (NT basal, PC control). Dataset 2: Cytotoxicity and metabolic activity of Gelidiopsis variabilis (GV) polysaccharide fractions (Funoran) in human keratinocytes (HaCaT) Description: This dataset comprises raw data from two independent experiments evaluating the effects of six Funoran fractions (GV-1A, GV-1B, GV-2A, GV-2B, GV-3A, and GV-3B) on immortalized human keratinocytes (HaCaT). HaCaT: MTT (0.50–0.01 µg/µL, 7 dilutions, 24 h; note: 1.00 µg/µL data also included) and alamarBlue (0.5 µg/µL, 24–72 h): normalized to NT, PC = 70% ethanol (MTT), NC = mitomycin C (alamarBlue). Statistics: Dunnett's (MTT) or Šídák's (alamarBlue) vs. NT. All data: mean ± SEM (n=4). Dataset 3: Cytotoxicity, metabolic activity, phagocytosis, and nitric oxide production of Gelidiopsis variabilis (GV) polysaccharide fractions (Funoran) in RAW264.7 macrophages Description: This dataset comprises raw data from four independent experiments evaluating the effects of six Funoran fractions (GV-1A, GV-1B, GV-2A, GV-2B, GV-3A, and GV-3B) on murine RAW264.7 macrophages. RAW264.7: MTT (0.50–0.01 µg/µL, 7 dilutions, 24 h): normalized to NT, PC = 70% ethanol, Dunnett's vs. NT (n=3–4). alamarBlue (0.5 µg/µL, 24–72 h): normalized to NT, PC = mitomycin C, NC = untreated, Šídák's vs. NT (n=4). Phagocytosis (0.5 µg/µL, 24 h): normalized to NT, PC = LPS, Dunnett's vs. NT (n=4). NO production (0.5 µg/µL, 24 h): expressed as µM, PC = LPS, Dunnett's vs. LPS (n=4). All data: mean ± SEM. Dataset 4: Cytotoxicity, metabolic activity, and barrier integrity effects of Gelidiopsis variabilis (GV) polysaccharide fractions (Funoran) in human colorectal adenocarcinoma cells (Caco-2) Description: This dataset comprises raw data from three independent experiments evaluating the effects of six Funoran fractions (GV-1A, GV-1B, GV-2A, GV-2B, GV-3A, and GV-3B) on human Caco-2 cells (colorectal adenocarcinoma model). Caco-2: MTT (0.50–0.0005 µg/µL, 11 dilutions, 24 h, down to 0.5 ng/µL): normalized to NT, PC = 70% ethanol, Dunnett's vs. NT. TEER (0.5 µg/µL, 0–48 h): Ω·cm², PC = 70% ethanol (disruption control), two-way ANOVA + Bonferroni's (each time point vs. baseline). All data: mean ± SEM (n=4).
