NMR analysis of two Tim10-mutants of TIM9·10. (A-D) F33QTim10. (E-H) V29KTim10. The BEST-TROSY 1H-15N NMR spectra (A,E) of the mutants (blue) are similar to those of wild-type TIM9·10 (black). (B, C, F, G) Residue-wise chemical-shift perturbations, calculated as the weighted sum of 1H and 15N chemical shifts. (D, H) Localization of residues with significant chemical-shift perturbations are shown in green and light blue, and the mutated residues, F33 and V29, are shown as dark blue sticks. Overall, chemical-shift perturbations are modest and located in the vicinity of the mutated residue, revealing that the mutants are structurally similar to the wild-type TIM9·10 complex.
