Steps to reproduce

We first phenotyped CB-SC, then we developed the MLR assays using enriched CD4 T cells and allogeneic MDDC. We ordered several leukpacks and isolated CD4 T cells and generated MDDCs to mix match for allogeneic MLR. CB-SC were provided by Dr. Zhao and we cultured in the lab from fresh buffy coats derived from Cord blood. We performed MLR in direct and indirect contact with CB-SC and also tested microvesicle enriched sups. Cytokine profiling was performed only on direct MLRs. The mouse study was performed last. We tried to harvest enough material for injecting microvesicle sups but there was not enough so we injected CBSC as the therapeutic agent, which worked for our purposes.
