Dataset for "Sex-dependent membranopathy in stored human red blood cells"

Description

This Dataset contains the results of analysis of cold-stored, human packed Red Blood Cells (pRBCs). The blood was withdrawn from men N=12, aged <30 years (N=3), 30-39 years (N=4), >40 years (N=5) and women N=12, aged <30 years (N=4), 30-39 years (N=4), >40 years (N= 4). The measurements were conducted weekly for 8 weeks. - 'Data for Figure 1' contains the results of biochemical analysis of (A) glucose, (B) lactate, (C) cholesterol, (D) triglycerides, (E) LDH, (F) free iron levels in pRBCs, performed with use of biochemical analyzer ABX Pentra 400 (Horiba Medical, Japan). - 'Dataset for Figure 2' contains the results of flow cytometry analysis of surface protein expression in pRBCs, namely CD47 and Phosphatidylserine, performed with use of LSRII flow cytometer (BD Biosciences). - 'Dataset for Figure 3' contains the results of flow cytometry analysis of Phosphatidylserine expression in pRBCs with regard to patients' age. - 'Dataset for Figure 4A' contains the results of ektacytometry analysis of pRBC deformability, perforemd with use of RheoScanAnD 300 (RheoMeditech, Seoul, Korea) . Elongation Index was calculated on the basic of the geometry of the elliptical diffraction pattern: (A-B)/(A+B), where A is light, and W is the width of the deformed cell. El values were calculated for shear stress at 20 Pa. - ' Dataset for Figure 4B' contains the results of complete blood count, comprising mean corpuscular volume (MCV), mean corpuscular hemoglobin concentration (MCHC) mean corpuscular hemoglobin (MCH), red blood cell count (RBC), hemoglobin (HGB) and hematocrit (HCT) performed using a hematology analyzer Abc Vet (Horiba Medical, France). - 'Dataset for Figure 5' contains the results of the atomic force microscopy (AFM) investigation of microparticle shedding from pRBCs, performed with use of WITec confocal CRM alpha 300 Raman microscope combined with an AFM (WITec, Ulm, Germany) . AFM images of 256 × 256 lines and 512 x 512 lines were collected in air conditions, from three different areas of 25 x 25 µm2, around ̴8 x 8 µm2 and 1.5 x 1.5 µm2. In order to determine the changes of RBCs surface and in situ formation of microparticles, cross sections of the AFM images were analyzed.
