Description

These experiments were carried out as part of our project on determining the effect of theta frequency sound that is used in hypnotherapy to facilitate theta state induction in brain.

2.2 Chronic Stress induction Chronic stress was induced in mice by exposing them to several stressors for 14 days on alternate days. Briefly, mice were acclimatized for one day prior to stress induction. Next day, mice were placed on orbital shaker at 50 rpm for 1 hour. On the alternate day, mice will be starved for 24 hours. On the next alternate day, mice were exposed to dry heat for 10 minutes followed by exposure to tilted cages the next alternate day. These stressors were repeated for the next week. 2.3 Theta sound exposure Mice werev divided in to following groups: mice with no stress and no sound treatment, mice without stress given sound treatment alone (in a sound proof chamber), mice with stress and no sound treatment, mice with stress and treated with theta sound (in a sound proof chamber), mice with stress treated with diazepam (1mg/Kg). Behavioral assessments and assays were conducted to all the groups. 2.3.1 Elevated plus maze test Animals were assessed for anxiety using elevated plus maze test. Briefly animals were place on a plus maze facing the closed arm. Animals will be allowed to explore the maze for 5 minutes and time spent in open arms will be recorded and analyzed. 2.3.2 Open Field Test Open field test were used to assess anxiety like behaviors after treatment with theta sound. Briefly, mice were placed in open field arena. They were allowed to explore the field for 1 minutes followed by recording of their activity for 5 minutes. The latency period and time spent exploring the arena were recorded.
