qPCR results of semi-continuous anaerobic digestion with and without vacuum stripping of digestate

Description

To avoid inhibition of ammonia on acetogenesis and methanogenesis, a small portion of in-reactor digestate can be replaced daily by vacuum stripped digestate. Vacuum stripping of ammonia may also increase methane generation. Key methanogens and syntrophic bacteria were identified by qPCR in semi-continuous anaerobic digesters with and without vacuum stripping of digestate.

Steps to reproduce

DNA was extracted with a DNeasy PowerSoil Kit (Qiagen, Germantown, MD, USA). The extracts were prepared at 20 ng total DNA per reaction mixture. Quantitative polymerase chain reaction (qPCR) was performed in 25 µL volumes on a QuantStudio 3 (Life Technologies).
