Data set for "Improved split TEV GPCR beta-arrestin recruitment assays via systematic analysis of signal peptide and a beta-arrestin binding motif variants" as published in Wu et al., Biosensors 2023, 13(1), 48; https://doi.org/10.3390/bios13010048 Data contain firefly luciferase readings from split TEV GPCR beta-arrestin-2 recruitment assays in four cell lines (HEK-293, U-2 OS, HeLa, and PC12-tetOff cells). Data from dose response assays for GLP1R and GCGR were performed in HEK-293 cells (data files "DRC").

Steps to reproduce

Plasmids encoding G protein-coupled receptors (GPCRs) linked to split TEV assay components were transiently transfected into HEK-293, U-2 OS, HeLa, and PC12-tetOff cells and stimulated with cognate ligands for 6h. Cells were lysed in Passive Lysis Buffer (Promega). Firefly luciferase activity was measured in a Mithras LB 940 Microplate Reader (Berthold Technologies) using the software MicroWin2000.
