High-throughput sequencing of single neuron projections reveals spatial organization in the olfactory cortex

Description

We mapped the brain-wide projection patterns of 5,309 olfactory bulb and 30,433 piriform cortex output neurons at single-cell resolution using barcode labelling and high-throughput sequencing based technologies (MAPseq and BARseq). Basically, we label individual neurons within olfactory bulb (OB) or piriform cortex (PC) by unique barcoded RNA, which can replicate and transport to axonal terminals. We then dissected targeted brain regions and injection sites, extracted RNA and sequenced for the barcodes as the readout of the brain-wide projections of barcoded neurons. Here we provided the images of dissected brain slices from 13 mouse brains: 8 brains for OB MAPseq/BARseq by Laser Capture Microdissection (LCM) and 5 brains for PC MAPseq by manual dissection with scalpel.
