Distribution of organohalide-respiring bacteria (OHRB) in chlorinated aliphatic hydrocarbons-contaminated site: Insights from redox zoning and functional genes-Chen

Description

1. Research Hypotheses OHRB distribution and activity in CAHs-contaminated sites are shaped by redox zonation (nitrate/iron/sulfate reduction and methanogenesis). The sulfate-reducing zone is the most active region for OHRB. OHRB and dechlorination genes (e.g., vcrA) exhibit synergistic coexistence. 2. Data Content This dataset includes: 16S rRNA sequencing data: Raw FASTQ files and OTU abundance tables. Functional prediction data: FAPROTAX-based functional classifications. qPCR data: Copy numbers of vcrA, entC, and phE genes. Environmental parameters: Depth, ORP, pH, temperature, CAHs concentrations. 3. Data Collection Sampling: Soil/groundwater samples collected along a 0–10 m vertical profile at a CAHs-contaminated site in Tianjin, China. Three replicates per depth. Sequencing: 16S rRNA V3–V4 region sequenced on Illumina NovaSeq; OTU clustering at 97% similarity against SILVA database. qPCR: SYBR Green assay with standard curves for absolute quantification. Functional prediction: FAPROTAX for metabolic function annotation. Statistics: SEM, PCoA, VPA, and co-occurrence network analysis in R. 4. Key Findings & Interpretation Diversity: Higher in shallow layers (<3 m); depth is the primary driver of community structure. Functional genes: vcrA peaked at 5 m; entC and phE showed depth-specific patterns. OHRB active zone: SEM confirmed the sulfate-reducing zone (5 m) as the most active region for OHRB (path coefficient > 0.6, p < 0.01). Synergistic coexistence: OHRB (Pseudomonas, Geobacter, Dehalobacter) co-occur with dechlorination genes, suggesting functional gene abundance as a potential bioindicator. Limitations: 16S rRNA provides relative abundance; FAPROTAX predictions are database-dependent. 5.
