CircRNA sequencing in hypertrophic myocardium from abdominal aortic coarctation surgical mice

Description

Pathological cardiac hypertrophy is a maladaptive cardiac remodeling in variable clinical diseases involving Hypertension, Cardiomyopathy and so on, which was still a major public healthcare problem with high morbidity and mortality in the worldwide.There is an unmet need to detect underlying heart failure sensitively and to reverse myocardial enlargement and fibrosis effectively. Several stimuli and signaling mechanisms have been postulated in pathological cardiac hypertrophy on genome and transcriptional levels, such as impaired Ca2+ handling, mitochondrial dysfunction,sm6A methyletation and so on. Researchers are devoting to find new targets to resolve the knotty matter and breakthroughs are made via next-generation sequencing, non-coding RNAs into researchers' fields of vision,and increasing mumber of circular RNA was detected in cardiac hypertrophy. We established pressure overloading-induced cardiac hypertrophy model in C57B/L mice at 8 weeks by AAC(abdominal aortic coarctation) surgery and ventricle myocardium were sent for cricRNA sequencing(both for 4 samples in sham and surgery group).We finally identified a total of 9780 circRNAs in two groups,among which 5 molecules were identified to have increased expression and 25 molecules to have decreased expression compare to sham group.
