Description

Data included below is related to the manuscript, "Repair of replication dependent double-strand breaks differs between the leading and lagging strands." Original images of Southern and western blots included in the publication are labeled with the corresponding figure numbers. A PowerPoint file is also included which indicates the cropped region used for the figures included in the manuscript. Primary outputs of the screen analysis used to generate figures are included in the folder labeled, "Results from the genome-wide screen." The "Combined Screen analysis" sheet includes data used to produce the Volcano plots in Figure 5 (mean_dif, and p columns). This sheet can be used to see growth differences after induction of nCas9 for all non-essential genes in the deletion library. The "Mean_growth_diff_by_trial" sheet includes the data used to produce the plots in Figure S5. For each strand_id (gene) and query_id (gRNA), the mean growth difference from both repeats of the screen are used to make the scatter plot displayed in Figure S5.
