Description

This is the raw data set used in the analysis from the manuscript "Liver vs. spleen: time course of organ-dependent immune gene expression in LPS-stimulated toads" by Felipe Floreste et al., published through DOI: This table contains values of immune proteins mRNA levels of Rhinella diptycha toads injected with LPS or saline and individually sampled 1h, 3h, 6h and 18h post-injection. Values are expressed as fold-change (FC) relative to the gene expression of housekeeping gene β-actin.

The mRNA level data (C1s, IFN-γ, IL-1β, IL-6, and IL-10) were initially submitted to the Shapiro-Wilk test of normality (n ≤ 50) and Levene’s test for homogeneity of variance. Since all variables showed an absence of normality or homogeneity of variance, they were transformed to Log10 to fit the assumptions of parametric tests. We used sets of mixed ANOVAs to investigate the effects of treatment and time in the cytokine expression levels. The mixed ANOVA allows the analyses of both between (independent) and within (repeated) subject variation in two or more groups. Protein mRNA levels (C1s, IFN-γ, IL-1β, IL-6, and IL-10) were considered as dependent variables, organ (spleen or liver) was considered as a within factor, and treatment (LPS or saline) and time (1h, 3h, 6h, and 18h) were treated as between factors. When appropriate, pair‐wised comparison tests with Bonferroni's adjustments were applied. Spearman non-parametric correlations were used to investigate possible relations amongst the variables within each treatment group.
