Steps to reproduce

Laboratory Animals Kunming mice (8 weeks, 19-20 g, female) had free access to sterile food and clean bottled water and were kept within a spacious cage at room temperature (25 ± 2 °C). The mice were intraperitoneally inoculated with T. gondii type II Prugniuad (Pru) tachyzoites, and brain cysts of the PRU strain were collected from the infected mice. Six domestic cats (Felis catus, Chinese Li Hua breed, 7-9 months) were randomly assigned to the experimental group (10_DPI) and control group (Control), with 3 cats in each group. All of the experimental cats were confirmed to be negative for T. gondii by using MAT, and no other major feline diseases were detected by ELISA. Each cat in the experimental group was intragastrically challenged with 600 tissue cysts diluted in 0.9% normal saline, while the cats in the control groups were treated with the same amount of saline. The cat's intestinal epithelial tissue was harvested by a cell scraper at 10 days post-infection (DPI), flash frozen in liquid nitrogen, and then sent to the company BGI (Shenzhen, China) for protein glycosylation analysis. Finally, the remaining laboratory animal tissues and T. gondii were subjected to sterilizing treatments, such as autoclaving. Confirmation of Infection Model Establishment Genomic DNA was extracted from the harvested samples by using a commercial kit (QIAGEN, Dusseldorf, Germany) according to the manufacturer’s instructions. PCR assay detection of T. gondii was performed by using the B1 gene as described previously. Parasites for histological observation were stained with H&E and observed by light microscopy (Olympus, BX41, Tokyo, Japan).
