The role of alcohol dehydrogenase 1C in regulating inflammatory responses in ulcerative colitis

Description

The online Gene Expression Omnibus (GEO) database was searched for the following microarray datasets: expression profiles containing colon tissue samples from patients with active ulcerative colitis (UC), normal controls, and of patients with UC who did not receive any treatment. Based on these criteria, three microarray datasets, GSE75214, GSE87466, and GSE92415 were selected. The GSE75214 gene expression dataset contained 74 active UC and 11 normal controls, which was in line with the platform GPL6244 (HuGene-1_0-st) Affymetrix Human Gene 1.0 ST Array. However, GSE87466 and GSE92415 gene expression profiles were matched with GPL13158 (HT_HG-U133_Plus_PM) Affymetrix HT HG-U133 array plus PM array plate. There were a total of 87 active UC and 21 normal controls in the GSE87466 dataset, and 53 active UC and 21 normal controls in GSE92415. To further identify potential differentially expressed genes (DEGs) in UC, the GEO2R online program based on the R software “limma” package was used to compare the active UC and normal groups. The thresholds for identifying DEGs were set as: adjusted P value < 0.05 (adj. P val < 0.05) and log2(fold change) > 1.0. A total of 1169 (DEGs are shown in "DEGs identified in GSE75214 dataset.docx"), 1389 (DEGs are shown in "DEGs identified in GSE87466 dataset.docx"), and 1306 DEGs (DEGs are shown in "DEGs identified in GSE92415 dataset.docx") were identified from the GSE75242, GSE87466, and GSE92415 datasets, respectively. Among them, 480 upregulated and 227 downregulated DEGs (DEGs are shown in "DEGs commonly changed in GSE75214, GSE87466 and GSE92415 datasets.docx") were common in all datasets.
