Description

Lipidomic abundance data (percent of total, percent of class) for 84 bulk patient tumors, 29 mouse xenografts, 43 gliomasphere cultures, and 27 biological replicates of genetically manipulated gliomasphere cultures. Further details on the data provided are described here https://youtu.be/PjUbVt968co Data related to Figure 1C including lipid species - gene correlation values and clusters can be found in the "Correlation Analysis" folder.

Steps to reproduce

Prior to extraction, a 13-class lipid class Lipidizer Internal Standard (AB Sciex, 5040156) was added to each sample. After two extractions, the pooled organic layers were dried down in a Genevac EZ-2 Elite, and lipid samples were resuspended in 1:1 methanol/dichloromethane with 10 mM Ammonium Acetate. Samples were analyzed on the Sciex Lipidyzer Platform for targeted quantitative measurement of 1100 lipid species across 13 classes. Differential Mobility Device on Lipidyzer was tuned with SelexION tuning kit (Sciex 5040141). Data analysis was performed according to the workflow published by Su, B. et al (2021).
