Description

Capsicum annuum (Bell pepper cv. California Wonder) and Capsicum chinense (Habanero pepper) plants were infected with Tomato brown rugose fruit virus (ToBRFV). In this repository, we showed some results. First, symptom development was compared between mock-inoculated and infected plants. Then, we performed droplet digital PCR (ddPCR) to quantify and compare the number of viral copies in systemic leaf samples. On the other hand, RTPCR was carried out with RNA samples of Opuntia ficus and Nicotiana tabacum infected with Opuntia Virus 2 (OV2) and Tobacco Mosaic Virus (TMV), respectively in order to test the specificity of the ToBRFV primers. These viruses were detected with specific primers for OV2 and TMV, demonstrating their suitability as negative controls for CP ToBRFV detection by ddPCR. The integrity and quality of biological material was tested by PCR amplifying ACTIN-11 and GAPDH fragments as endogenous genes.
