RGB values of B-10 milk biofilms on stainless steel coupons

Description

The fluorescence micrographs of EtBr-stained B-10 milk biofilms on stainless steel coupons (SSCs) were analyzed using the ImageJ 1.52a software. The RGB values obtained were in decreasing manner when comparing before and after the immersion in 100 mg/ml of P-03 (an unknown species of Pestalotiopsis) culture crude extract at 50 degrees Celsius for 15 min followed by EtBr (ethidium bromide) staining. The results suggest that the recorded intensity (CPS) of light emitted by the EtBr stained the extracellular matrix (ECM), which covered the B-10 milk biofilms, was disrupted. The values also showed in the 3-D plot surface generated by the software. ECM degradation is one of the key to biofilm control. The P-03 crude extract has antimicrobial property with MIC of 6.25 mg/ml. Degrading the ECM will kill the exposed B. cereus s.l. biofilm cells, but not the spores, by the fungal crude extract and therefore a possible alternative antibiofilm agent.
