Steps to reproduce
We generated mice with a targeted deletion of IKKα in the DC lineage using Cre-loxp system. FACS was performed to assess IKKα modification on DC, T cell and other immune cells. The level of cytokines was assessed by ELISA and CBA assay. Signaling pathway altered by IKKα knock out were identified by RNA-seq and RT-qPCR. Tumor xenograft model were used to estimate DC specific depletion of IKKα in primary response and tumor growth. The mechanism of IKKα regulatory to DCs migration and maturation were analyzed by Transwell, FACS and western blot.
