Description

Dataset accompanying the 2026 article: Hypothalamic amyloid levels are associated with early sex-dependent alterations in energy homeostasis in TgF344-AD rats. We hypothesized a bi-directional relationship between peripheral metabolic impairments and Alzheimer's disease neuropathology. This dataset includes pre and post-mortem measurements from male and female Tgf344-AD rats. We detected soluble amyloid beta 40 and 42 in the hypothalamus of TgF344-AD rats alongside sex-dependent impairments in peripheral metabolic indices. Amyloid beta 42 positively correlates with separate impaired measures in male versus female TgF344-AD rats. These findings suggest that Alzheimer's disease pathology in the hypothalamus may induce or contribute to peripheral metabolic impairments in a sex-dependent fashion which can then exacerbate AD pathology. Steps to reproduce

See publication in press for extended methods. TgF344-AD rats were assayed either on a chow diet or a HFHS diet consisting of 45% calories from fat and 17% from sucrose. This diet was maintained for 11 weeks. Weight was measured weekly. Body composition was measured after 10 weeks of diet using an EchoMRI. Peripheral blood glucose clearance was measured by administration of an IP bolus of glucose followed by blood measurements for 120 minutes following injection. Body temperature was measured using surgically implanted temperature monitors. Feeding was measured using automatic balances. Various organs and peripheral fat pads were collected, alongside brain tissue. Western blots were utilized to measure UCP1 content of Brown adipose tissue and ELISA was used to measure soluble human amyloid beta 40 and 42 in the hypothalamus and soluble human amyloid beta 42 in the cortex. Data were analyzed using R and GraphPad prism.
