Steps to reproduce

Mice were transported from their home cages to the behavioral testing room and allowed 30 min to acclimate before the start of each test. All behavioral procedures were performed during the evening light cycle and scored by an experimenter who was blinded to genotype or treatment. Y maze test. This behavior mainly tests the working memory of mice88. The Y-maze was made of black plastic. The maze had three arms (ABC), each 50 cm long, 20 cm high, and 10 cm wide at the bottom and top. The behavior of mice was video recorded using Noldus EthoVision XT (Ugo Basile). At the beginning of each recording, individual mice were placed at the end of arm A and allowed to freely explore the maze for 8 min. Spontaneous alternation (%) was defined as consecutive entries into three different arms (ABC) divided by the number of possible alternations (total arm entries - 2). A higher percentage of spontaneous alternation behavior indicates better cognitive ability. MWM test. This behavioral test mainly tests the spatial learning and memory of mice89. It was conducted according to the previous protocol. In brief, the test was conducted in a dark circular pool (1.2 m diameter, 0.5 m height) filled with opaque white water (22 ± 1℃) and 35 cm deep. Reference cues of different shapes were placed along the walls around the tank. A fixed escape platform (10 cm diameter) was placed at the midpoint of a quadrant, hidden 1.5 cm below the water surface. During the training period, each mouse performed 6 trials per day for 7 consecutive days. Each trial started from a different position according to the semi-random sequence distribution determined by Vorhees and Williams. The mouse faced the pool wall when entering the water. The mouse was allowed to swim freely for 60 s to find the platform. Mice that failed to find the platform within 60 s were guided to the platform and stayed there for 20 s. The latency was the average of four training sessions. The memory test (probe test) was performed 24 h after the last training. The platform was removed and each mouse was allowed to swim freely for 60 s. The time it took for the mouse to reach the platform area and the number of times the mouse crossed the platform position were recorded. The swimming performance of each mouse was recorded and analyzed using Ethovision video tracking software (Noldus, The Netherlands).
