Fresh skin biopsies were placed in saline at 4°C until further processing. The epidermis was separated from the dermis through Dispase II digestion at 4°C for overnight. Dermis single-cell suspensions are subjected to FACS to exclude doublets, debris, DAPI-positive dead cells, from which CD45+ cells were sorted. Sorted cells were centrifuged and resuspended in 0.04% BSA in PBS. Cells were subjected for 10X single-cell sequencing.
