The genotypes of green frogs from water bodies of Western Ukraine

The file contains genotype data of 91 individuals of green frogs, collected in water bodies of Western Ukraine. Seven microsatellite loci were used: Rrid059А, Rrid082А, Rrid171, RlCA1b5, Res16, Res22, RlCA19

Steps to reproduce

DNA was extracted from samples of muscle tissue stored in 70% alcohol and also from buccal swabs according to the N. Pindasie et al. method. For the extraction of DNA, we used universal laboratory sets “Diatom™ DNA Prep” produced by the «Laboratoriya Isogen». For the PCR we used already prepared mixes of reagents specially developed for the PCR-amplification – GenePak® PCR Core produced by «Laboratoriya Isogen». Electrophoretic separation of obtained DNA samples was performed in 6% polyacrylamide gel with the usage of Tris-Borate-EDTA buffer (TBE Buffer). To estimate the length of fragments, a DNA plasmid pBR322 of E. coli prepared by HpaII restriction endonuclease was added to each of the gel plates.
