Description

Images correspond to the original, unprocessed versions that appear in Fig. S1A-B. BF in the name indicates transmission images, and Ste5-YFP indicates images acquired using the YFP filter settings. Movies correspond to the confocal whole-field images from which the cells that appear in Fig S2D were selected. There are 3 pairs of hyperstacks (three TFP: transmission; three YFP). One pair is for WT cells, the other delta-far1, and the other delta-fus3. Each hyperstack consists of 5 z-planes plus a sixth plane added at the end that corresponds to the maximal projection of the previous 5 stacks, and 41 time points. The time points (in minutes), relative to the addition of 1 micromolar alpha factor, are as follows: -2, -1, 1, 2.5, 4, 10, 15, 20. Thereafter, every 5 minutes. Note: images and hyperstacks are in 16-bit format. To be able to see the signal, we recommend using a compatible image viewer, such as ImageJ or Fiji.
