we compared the quality of bull frozen-thawed sexed semen after sexing sperm by cytotoxicity reaction to that of commercial sex-sorted semen using computer-assisted sperm analysis and imaging flow cytometry.

Steps to reproduce

The objective was to evaluate the quality of bull frozen-thawed sexed sperm after sexing by cytotoxicity reaction and commercial sex-sorted semen. Ejaculates were collected from the six bulls using an artificial vagina twice a week. Semen samples were divided into two group including conventional sperm bull (CON) and sexed semen by cytotoxicity reaction (Cy-SEXED). In addition, commercial sex-sorted sperm (SORTED) was compared in this study. Three groups were evaluated sperm motility and kinematics using computer assisted sperm analysis (CASA). Imaging flow cytometer were analyzed; sperm plasma membrane integrity was assessed using SYBR-14/PI, acrosome integrity and viability was assessed using Alexa Fluor®/PI, separation of X- and Y-chromosome-bearing sperm was assessed using hoechst 33342.
