Microsatellite genotype dataset of Plathymenia reticulata from native populations and seed lots in Brazil

This dataset contains multilocus microsatellite (SSR) genotype data for Plathymenia reticulata Benth., a tropical tree species native to the Atlantic Forest and Cerrado biomes of Brazil. The dataset comprises individuals sampled from eight native populations and six seed lots used in ecological restoration programs. Genotypes are reported as allele sizes (base pairs, bp) for each microsatellite locus. Each row corresponds to a single individual, and each locus is represented by two columns corresponding to the two alleles. These data were generated to evaluate the genetic diversity captured in seed lots relative to native populations. The dataset was used to estimate genetic diversity, observed and expected heterozygosity, allelic richness, inbreeding coefficient, genetic structure, relatedness, and recent bottlenecks. It supports the findings reported in the manuscript "Alleles, Seeds and Forests: Genetic Diversity of Seed Lots and Native Populations of a Tropical Tree in Support of Ecosystem Restoration Efforts." These data provide an important resource for conservation genetics and ecological restoration by documenting the genetic diversity represented in native populations and seed lots of Plathymenia reticulata. The dataset supports the development of evidence-based seed collection strategies aimed at maximizing genetic diversity, minimizing relatedness among seedlings, and improving the long-term resilience and adaptive potential of restored tropical forest populations. Steps to reproduce
The dataset contains multilocus microsatellite genotypes of Plathymenia reticulata. Each row represents one individual, and each microsatellite locus is represented by two columns corresponding to the two alleles scored as fragment sizes (bp). These data were used for population genetic analyses, including estimates of genetic diversity, inbreeding, genetic differentiation, relatedness, genetic structure, and recent bottlenecks, as described in the associated manuscript. Seed lots of P. reticulata were sampled by acquiring seeds from commercial nurseries located throughout the species’ natural distribution range. The seeds originated from six provenances across three Brazilian states: Goiás (1), Minas Gerais (3), and São Paulo (2).To investigate the genetic diversity in seed lots in comparison with natural populations, we sampled eight P. reticulata populations in the state of Bahia, Brazil, from forest remnants (NP1, NP2, NP3 and NP4) and savanna phytophysiognomies (NP5, NP6, NP7 and NP8). In each population (NP2, NP3, NP4, NP5, NP6, NP7 and NP8), an average of 26 adult individuals were sampled. The individuals were georeferenced, and young leaves were collected for analysis. The seeds underwent dormancy breaking through mechanical scarification using sandpaper applied to the lateral region near the hilum. Subsequently, they were placed in plastic trays containing washed sand and watered twice daily. Emerged seedlings were collected, stored in kraft paper envelopes, placed in plastic bags with silica gel, and kept at -20 ºC. A total of 24 seedlings were sampled for provenances, except for SL5, from which 19 seedlings were sampled. Genomic DNA was extracted from emerging seedling tissues and leaf samples collected from adult individuals using the Doyle and Doyle (1987) protocol. Samples were genotyped for nine nuclear microsatellite loci (SSR—Simple Sequence Repeats) following Oliveira et al. (2012). PCR products were analyzed via capillary electrophoresis using a Genetic Analyzer ABI/Hitachi 3500 (Applied Biosystems, USA) in a multiload system. Electropherogram analysis was conducted using GeneMarker software version 1.95 (SoftGenetics, State College, PA, USA).
