  • MARCKSL1 regulates spine formation and controls anxiety
    Abnormalities in the limbic neural circuits have been implicated in the onset of anxiety disorders. However, the molecular pathogenesis underlying anxiety disorders remains poorly elucidated. Here, we demonstrate that myristoylated alanine-rich C-kinase substrate like 1 (MARCKSL1) regulates the amygdala circuitry to control the activity of hypothalamic-pituitary-adrenal (HPA) axis and induces anxiety behaviors in mice. MARCKSL1 expression was predominantly localized in the prefrontal cortex (PFC), hypothalamus, hippocampus, and amygdala of the adult mouse brain. MARCKSL1 transgenic (Tg) mice exhibited anxiety-like behaviors dependent on corticotropin releasing hormone. MARCKSL1 increased the spine formation in the central amygdala, and the downregulation of MARCKSL1 in the amygdala normalized both increased activity of the HPA axis and elevated anxiety-like behaviors in Tg mice. Furthermore, there was increased MARCKSL1 expression in the PFC and amygdala in a brain injury model associated with anxiety-like behaviors. Our findings suggest that MARCKSL1 expression in the amygdala plays an important role in anxiety-related behaviors.

  • Replication Data for: Intestinal Disturbances Associated with Mortality of Children with Complicated Severe Malnutrition

    This is data is from a case-control study nested within the F75 intervention trial. By comparing the fecal metabolomic profiles and enteropathy markers between cases (children who died, n=68) and controls (children who were discharged alive, n=68), this study aimed to understand the association between intestinal disturbances wnad mortality among children with severe malnutrition admitted to hospitals in Kenya and Malawi. Targeted fecal metabolomics were performed using nuclear magnetic resonance spectroscopy. A total 68 metabolites were targeted. Four enteropathy markers, myeloperoxidase, calprotectin, alpha-1-antitrypsin, and intestinal fatty acid binding protein were quantified by ELISA. Lyophilization was used to determine water content of fecal samples.

  • Effects of BCG vaccination on donor unrestricted T cells in two prospective cohort studies
    The data were generated to address whether BCG vaccination can modulate donor unrestricted T cells by performing immunological analyses using flow cytometry on PBMC samples collected from infants and adults in clinical studies completed previously (doi: 10.4049/jimmunol.1501996). The results are published in Gela (2021) - Effects of BCG vaccination on donor unrestricted T cells in humans. `data_tidy_gela_durt.zip` is the source code to compile the R package `DataTidyGelaDURT`, which provides the following datasets: - data_tidy_freq_ifng: Background-subtracted frequencies of IFNg+ cells for various subsets.- data_tidy_freq: Frequencies of major cell populations- data_tidy_hladr_adult: HLADR mean fluorescent intensity (MFI) by cell type for infants.- data_tidy_hladr_infant: HLADR mean fluorescent intensity (MFI) by cell type for infants.- data_tidy_mem: Frequency of memory phenotype relative to the parent population. - Note that the package is also available at https://github.com/SATVILab/DataTidyGelaDURT (where any issues can be raised as well).
