Data and code for controlled iodine-125 seed displacement in prostate brachytherapy
Description
This dataset contains the input files, paired dose-volume data, and analysis code accompanying the manuscript “Impact of controlled iodine-125 seed displacement on prostate brachytherapy dose-volume metrics”. The study evaluated a fixed 39-seed iodine-125 computational prostate implant using OpenGATE 10.1.0 and Geant4 11.4.0 with an explicit Best Medical 2301 source model in homogeneous water. Fifteen independent displacement-direction families were evaluated at 1, 2, 3, and 5 mm. Each perturbed configuration was compared with a family-specific nominal control at 500,000 primary histories per seed. The archive includes nominal and perturbed seed coordinates, displacement unit vectors, source geometry and spectrum definitions, simulation parameters, absolute-dose calibration records, 75 generated OpenGATE input scripts, validation summaries, the final 60-record paired-effect dataset, statistical output tables, and analysis and plotting scripts. The primary endpoints are percentage changes in D90 and D95 and percentage-point changes in V100 and V150. D100 is included as an exploratory endpoint. The supplied paired-effect data and analysis scripts support reproduction of the reported statistical analyses and figures. The original computational DICOM/RTSTRUCT phantom and target mask are not included. Full voxel-level regeneration of dose-volume histograms additionally requires the original target geometry. These data describe sensitivity within a single computational implant and should not be interpreted as clinical localization tolerances or action levels.

Steps to reproduce
Extract the archive and open the Supplementary_Data_1 directory. Install the Python dependencies listed in requirements.txt. To reproduce the statistical analyses and endpoint figures, run: python analysis/final_statistical_analysis.py data/perturbation_production_v26_final_paired_effects_long.csv --outdir reproduced_analysis The analysis/figure1_study_design.py script reproduces the study-design figure. The opengate/generated_inputs directory contains the 75 final transport input scripts. Transport calculations require a compatible OpenGATE environment. Full voxel-level DVH regeneration also requires the original computational target mask, which is not distributed in this archive.
