Description

This dataset contains flow cytometry standard (FCS) files from individual 6–8-week-old wild-type C57BL/6J mice treated intraperitoneally with PBS or 50 µg high-molecular-weight polyinosinic-polycytidylic acid (poly I:C) 12 hours before analysis. Samples were collected across three independent experiments and included both male and female mice. Each FCS file represents one individual mouse, and samples were not pooled. All samples were acquired on a BD Fortessa (BD) . Compensation was generated using previously established single-color controls. The resulting compensation matrices are embedded in the FCS files.

Steps to reproduce

To reproduce the analysis, open the FCS files in FlowJo 10 or compatible flow cytometry analysis software. Use the compensation matrices embedded in the FCS files. Gate sequentially on singlets, CD45+ cells, Lineage- (CD3, CD19, B220, and NK1.1), CD64-F4/80- cells, CD11c+I-Ab+ cells, and then XCR1+CD172a- cDC1s. Within the total splenic cDC1 population, quantify the frequencies of BTLA-high CCR7-low cDC1s and BTLA-low CCR7-high cDC1s for each individual mouse.
