Effects of Tong-Xie-Yao-Fang (TXYF) Disassembled Recipes on Brain-gut Peptide in Diarrhea-type Irritable Bowel Syndrome(IBS-D) Rats

Tong-Xie-Yao-Fang was divided into white peony root-- divaricate saposhnikovia root(B-F) and dried tangerine peel-- largehead atractylodes rhizome(C-B) to treat IBS-D model rats.The data include Body weight, AWR score, Bristol stool sale, the expressions of BDNF, SP and CGRP in colon and hippocampus of all rats. B-F in TXYF could bi-directionally regulate the expression of BDNF, which is down regulating the expression of BDNF in colon tissue and up regulating the expression of BDNF in hippocampus tissue. B-F group could significantly down-regulate the expressions of BDNF, SP and CGRP in colon tissue.

Steps to reproduce

Stool characteristics: each rat was fed in a single cage 1 day before modeling, before treatment and after treatment. Feces were collected and observed for 4 hours. The collected feces were graded according to the Bristol stool sale. Abdominal wall reflex (AWR) scores: it was tested before treatment and after treatment. The rats only drank water 24 hours before the test. After smearing the self-made balloon with paraffin oil and stuffed it into the anus for about 7 cm, then slowly pressurized the balloon with a sphygmomanometer to observe the rats' response to balloon expansion stimulation under four pressure gradients of 20, 40, 60 and 80mmHg. The average value of AWR score at each pressure gradient was taken for 3 times. Two workers who did not know the grouping of the experiment were measured together, and the interval between each measurement was about 5 minutes. The expressions of BDNF, SP and CGRP in colon and hippocampus were detected by ELISA: 18 hours after treatment, 2% Pentobarbital Sodium(2.5ml/kg) was injected intraperitoneally to anesthetize, and the complete colon slice was taken out with a length of about 2cm. At the same time, the rat skull was quickly opened with biting forceps and the rat hippocampal tissue was completely removed. The obtained colonic and hippocampal tissues were quickly transferred to the refrigerator at - 80 ℃ for storage. According to the instructions of the kit, the OD value at 450nm was measured by microplate reader and the contents of BDNF, SP and CGRP in rat colon and hippocampus were calculated respectively.
