Steps to reproduce
1) A poly(A) mRNA was isolated from 150 mg of saline-washed hemocytes (crude weight) using a SileksMag-oligo(dT)30 kit (Sileks, Russia) following the manufacturer's protocol. 2) The amount of mRNA obtained was 1.1 μg, as determined by a NanoDrop Ultra spectrophotometer (Thermo Fisher Scientific, Waltham, MA, USA). 3) The isolated mRNA was then used for cDNA synthesis with a Roche cDNA synthesis system. 4) The cDNA was sequenced using an Ion Torrent device, and a cDNA library was created using Mira Assembler version 2.0. The resulting library contains 1,064,218 transcripts.
