In this study, we identified PP2A and CDK16 as regulators of WIPI2B phosphorylation and neuronal autophagosome biogenesis. To do so, we utilized a C. elegans reverse genetics screen and a visual readout of autophagy function to identify regulators of neuronal autophagy. We verified activity of PP2A and CDK16 on mammalian WIPI2B through in vitro kinase and phosphatase assays. Additionally, we showed that manipulating PP2A and CDK16 expression in primary murine neurons (DRGs) alters WIPI2B puncta formation and rates of autophagosome biogenesis. These data include western blots and fluorescent micrographs of C. elegans neurons and primary murine neurons.
