Antioxidant and immunostimulant activities of Chilean seaweed polysaccharides

This dataset contains experimental raw data evaluating the relationship between chemical structure, molecular weight (MW), and biological activities of soluble polysaccharides extracted from three native Chilean seaweed species: the brown kelps Lessonia berteroana and Lessonia trabeculata, and the red macroalga Sarcodiotheca gaudichaudii.

Steps to reproduce
The experimental workflow was executed as follows: • Polysaccharide Extraction & Purification: Native sodium alginates were extracted from Lessonia stipes using alkaline extraction followed by calcium precipitation. Native carrageenans were isolated from S. gaudichaudii via hot aqueous extraction. All residues were purified through exhaustive dialysis. • Oxidative Depolymerization: To study the influence of molecular weight on bioactivity, a fraction of each native polysaccharide was subjected to free-radical depolymerization using 9% hydrogen peroxide (H2O2) at pH 7.5 and 60°C for 3 hours. • Chemical Characterization: Native and depolymerized fragments were profiled using Fourier Transform Infrared (FTIR) spectroscopy, Gel Permeation Chromatography (GPC) for weight-average molecular weight (Mw) determination, colorimetric assays (total sugars, sulfates, 3,6-anhydrogalactose), and elemental CHNS analysis. • Bioassays: 1. Antioxidant Activity: Evaluated via 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assays across a dose-dependent range (0.16 to 10.0 mg mL-1). 2. In Vitro Macrophage Assays: Cytotoxicity and cell viability were verified on the RAW 264.7 murine macrophage cell line using MTT assays. Immunostimulatory potential was assessed by exposing non-cytotoxic concentrations (5 to 200 µg mL-1) of the polymers to the cells and quantifying the release of pro-inflammatory cytokines (TNF-α and IL-6) through Uncoated ELISA Kits .
