Confocal images of TCS::GFP in wt and ahp4 mutants

Description

This contains data taken by Elina Chrysanthou of the TCS marker in ahp4 and wild-type Arabidopsis plants. The data set was generated to test if there was cytokinin response was altered in the phloem companion cells of the mutant. The findings are published in Chrysanthou et al., 2025. All plants have been treated with 300 mM mannitol for 24h. Data series used for analysis consist of xzy stacks of 20 images. The folders are based on the time the images were taken. The marker was introduced to the ahp4-1 mutant by crossing and segregating lines genotyped. The files with the homozygous mutants are marked mutant and the wt plants that segregate are marked WT. Images are saved in .lif format and can be opened using either Leica's confocal software or using the Bioformats plugin for imageJ.

Steps to reproduce

Plants were grown on agar plates for 5 days then transferred to 300 mM mannitol plates for a further 24h. GFP was imaged with a 488 laser and tDtomato (marking the membranes) with a 563 laser. All imaging was performed on a Leica SP8 microscope.
