Serum interleukin-10 in preeclamptic and normotensive pregnancies among Javanese and Batak women, Medan, Indonesia

This dataset contains de-identified participant-level data from a case–control study comparing serum interleukin-10 (IL-10) concentrations between pregnant women with preeclampsia and normotensive pregnant women, and between the two largest ethnic groups in Medan, North Sumatra, Indonesia (Batak and Javanese). Sixty-eight pregnant women beyond 20 weeks of gestation were recruited by consecutive sampling at Prof. Chairuddin P. Lubis Hospital, Universitas Sumatera Utara: 30 women with preeclampsia and 38 normotensive controls, with ethnicity balanced within each group (34 Batak, 34 Javanese in total). Serum IL-10 was measured by enzyme-linked immunosorbent assay at the Integrated Laboratory, Universitas Sumatera Utara. Recorded variables are study group, ethnicity, maternal age, gestational age at sampling, parity, highest level of education, body mass index category, urine dipstick proteinuria, and IL-10 concentration in pg/mL. All direct identifiers were removed before deposit: participant names were deleted and replaced with sequential anonymous identifiers, and no dates of birth, admission dates, medical record numbers or addresses are included. The dataset is complete, with no missing values. It is provided so that the analyses reported in the associated article — group comparisons, ethnicity-stratified comparisons, multivariable logistic regression, and receiver operating characteristic analysis — can be reproduced and re-examined.

Steps to reproduce
Study design and setting. Observational analytic case-control study at Prof. Chairuddin P. Lubis Hospital, Universitas Sumatera Utara, Medan, North Sumatra, Indonesia, with serum assays at the university's Integrated Laboratory. Recruitment ran from April 2025, after approval by the Health Research Ethics Committee, Universitas Sumatera Utara, until the target sample size was reached. Participants. Sixty-eight pregnant Batak or Javanese women beyond 20 weeks of gestation were enrolled by consecutive sampling: 30 with preeclampsia and 38 normotensive controls, ethnicity balanced within each group. Preeclampsia was diagnosed as new-onset blood pressure >=140/90 mmHg on two occasions at least 15 minutes apart after 20 weeks of gestation, with significant proteinuria or, in its absence, evidence of maternal end-organ or uteroplacental dysfunction. Controls had blood pressure below 140/90 mmHg and no proteinuria. Ethnicity was assigned only when both parents belonged to the same ethnic group, as reported by the participant. Exclusions: multiple pregnancy, diabetes mellitus, cardiac disease, active infectious disease, renal disease, autoimmune disorder, chorioamnionitis, or an unusable sample. Minimum sample size (14 per group) was calculated with the two-means formula at alpha 0.05 and 90% power. Measurements. After written informed consent, maternal age, gestational age, parity, medical and drug history, weight and height were recorded. Body mass index was classified by Asia-Pacific criteria (normal 18.5-22.9; overweight 23.0-24.9; obese >=25.0 kg/m2). Blood pressure was measured with an appropriately sized cuff and urine tested for protein by dipstick. Venous blood (5 mL) was drawn from the median cubital vein into EDTA tubes by trained staff. Serum interleukin-10 was measured by enzyme-linked immunosorbent assay (ELISA), with samples from both groups processed in the same runs and laboratory personnel unaware of group allocation. Data processing. Before deposit, participant names were deleted and replaced with sequential anonymous identifiers; no dates of birth, admission dates, medical record numbers or addresses are included. Variable names and category labels were translated from Indonesian into English, decimal commas converted to decimal points, and one gestational-age entry recorded as "36=37" corrected to "36-37". No values were imputed, removed or otherwise altered; the dataset is complete. Analysis. Normality was assessed with the Kolmogorov-Smirnov test. Groups were compared with the Mann-Whitney U test. Binary logistic regression estimated the association between IL-10 and preeclampsia, unadjusted and adjusted for maternal age, obesity and nulliparity. Receiver operating characteristic analysis assessed discrimination, with a bootstrap confidence interval for the area under the curve (2000 resamples). Software: SPSS v22 and Python 3.11 (statsmodels, scikit-learn). Two-sided p<0.05 was considered significant.
