HPLC-ESI-MS characterization of phytochemicals in Sarghina extract

Description

HPLC analysis of sarghina extract was performed on the Dionex Ultimate 3000 HPLC System (Thermo Fisher Scientific, USA), equipped with the Ultimate 3000 RS Variable Wavelength UV DAD. DAD was performed in-line in UV–VIS spectra in the range of 200–800 nm. A column (Acclaim Polar Advantage 5µm - C18 - 250 mm x 4.6 mm) was used and the oven temperature was maintained at 30°C. Data acquisition was performed by Xcalibur software (version 3.1, Thermo Fisher Scientific).

Steps to reproduce

The phase mobile consists of two solvents: 0.1% formic acid in water (Solvent A) and 0.25% acetonitrile-formic acid (60:40 v/v) (Solvent B). The following gradient program was run: 0 min, 100% A; 15 min, 80% A; 30 min, 70% A; 50 min, 100% B. The flow rate of the mobile phase was 1.3 mL/min. The extract was subjected from HPLC to the Mass Spectrometer System (Tribrid Thermo Scientific Orbitrap ID-X Tribrid). Temperature and capillary voltage were 300 ° C and 70 V, respectively. Full scan spectra were acquired in the range 100 to 1100 m/z in negative mode.
