Description

The Glue Grant 2 transcriptomic cohort is a multicenter prospective observational cohort of critically injured trauma patients enrolled from seven tertiary care trauma centers in the United States. The cohort includes 243 adult trauma patients with severe injury, defined by an Injury Severity Score (ISS) ≥16 and requirement for intensive care unit admission, together with 57 age- and sex-matched healthy volunteers as controls. Participating centers included the University of Florida, University of Pittsburgh Medical Center, University of Washington, University of California San Francisco, The University of Texas Southwestern Medical Center, Massachusetts General Hospital, and the University of Colorado Anschutz Medical Campus. Peripheral blood samples were collected longitudinally at predefined post-injury time points, including early injury, intermediate recovery phases, and recovery or hospital discharge when available. Transcriptomic profiling was performed on circulating immune-cell populations and linked to prospectively collected clinical metadata, including ICU length of stay, organ failure, survival status, and recovery-related outcomes. In this study, the Glue Grant 2 cohort was used to develop and evaluate immune recovery clock models and to investigate the relationship between post-trauma immune transcriptional dynamics and clinical outcomes.

Steps to reproduce

Total RNA from isolated monocytes, T cells and neutrophils was processed for GeneChip-based transcriptomic profiling. Amplified cDNA was generated from total RNA using first-strand cDNA synthesis, second-strand cDNA synthesis, purification, SPIA linear isothermal amplification, fragmentation and biotin labeling. Fragmented and labeled cDNA was hybridized to Affymetrix GeneChip arrays at 45 °C for 18 h, followed by washing, staining, and scanning according to the GeneChip Expression Analysis protocol.
