In the study exploring the therapeutic effect of PPA on ovalbumin-induced allergic obese asthmatic mice, 16S rRNA sequencing was adopted to analyze the composition of gut microbiota.

Steps to reproduce

Genomic DNA was isolated from mouse feces using the MagBeams FastDNA Soil Kit (MP Biomedicines, CA, USA). PCR amplification of the V3-V4 hypervariable region within the 16S rRNA gene was performed using site-specific primers 338F (5 '- barcode+ATCCTACGGGAGGCAGCA-3') and 806R (5 '- GATACHVGGGTWTCTAAT-3'). After PCR amplification, sequencing reactions were performed using the NovaSeq 6000 SP kit on the Illumina NovaSeq sequencing platform. Using Vsearch for unit of classification (OUT) clustering operation, the similarity level is 97%.
